The mammalian target of rapamycin mTOR, also know as FRAP or RAFT, is a serine/ threonine protein kinase.
mTOR acts as a sensor for ATP and amino acids, balancing the availability of nutrients and cell growth.
When sufficient nutrients are available, mediated by phosphatidic acid, mTOR transmits a positive signal to p70 S6 kinase and participates in the inactivation of the eIF4E inhibitor 4E-BP1.
These events result in activation of the translation of specific mRNA subpopulations.
mTOR is phosphorylated at serine 2448 via the PI3 kinase/Akt signaling pathway and autophosphorylated at serine 2481.
Applications:Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight
Immunoprecipitation: 1:100Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage, store at -20°C.
Stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
