Lck, a member of the Src family of protein tyrosine kinases, is essential for T-lymphocyte activation and differentiation (1,2).
It is associated with a variety of cell surface receptors and plays a key role in T-cell antigen receptor linked signal transduction pathways.
The activity of Lck is regulated by protein kinases and phosphatases.
Phosphorylation of the C-terminal tyrosine 505 serves to downregulate Lck catalytic activity, while phosphorylation at tyrosine 394 leads to an increase in Lck activity (3,4).
The protein tyrosine phosphatase SHP-1 has been shown to dephosphorylate Lck at tyrosine 394 (5).
