Epidermal growth factor (EGF) receptor is a 170kD single-pass transmembrane tyrosine kinase.
Ligand binding to this receptor results in receptor dimerization, autophosphorylation (in trans), activation of various downstream signaling molecules and lysosomal degradation (1,2).
Tyrosine 845 resides in the activation loop of the receptor kinase domain (3,4).
Phosphorylation of Tyr845 may stabilize the activation loop, maintain the enzyme in an active state and provide a binding surface for substrate proteins.
It has been shown that c-Src is involved in phosphorylation of the EGF receptor on Tyr845, which is associated with modulation of receptor function (5).
Phospho-tyrosine 992 of activated EGF receptor is a direct binding site for the phospholipase C-gamma (PLCgamma) SH2 domain (6).
This binding results in activation of PLC-gamma-mediated downstream signaling.
Phosphorylation of Tyr1045 creates a major docking site for c-Cbl (7).
Binding of c-Cbl to the activated EGF receptor leads to assembly of ubiquitination machinery to the receptor, and enables receptor ubiquitination and degradation (8).
Phospho-tyrosine 1068 of activated EGF receptor is a direct binding site for the Grb2/SH2 domain (9).
This binding results in Ras activation through a Grb2/Sos-1 signaling mechanism (2).
Applications:Suitable for use in ELISA and Western Blot.

Recommended Dilution:Western Blot: 1:1000, incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight
