Caspase-9 (ICE-LAP6, Mch6) is an important member of the cysteine aspartic acid protease (caspase) family (1,2).
Upon apoptotic stimulation, cytochrome c released from mitochondria associates with procaspase-9 (47kD)/Apaf 1.
This complex processes procaspase-9 into a large active subunit (35kD or 17kD) and a small subunit (10kD) by self-cleavage at Asp315 (3–5).
Cleaved caspase-9 further processes other caspase members, including caspase- 3 and caspase-7, to initiate a caspase cascade, leading to programmed cell death (6–9).
In addition to self-cleavage, procaspase-9 also can be cleaved in vivo by caspase-3 at Asp330.
This process serves as positive feedback to amplify the apoptotic signal in the caspase activation pathway (3–5).
Applications:Suitable for use in ELISA and Western Blot.
