特徴

• Bad is a proapoptotic member of the Bcl-2 family that can displace Bax from binding to Bcl-2 and Bcl-xL, resulting in cell death.
• Survival factors such as IL-3 can inhibit the apoptotic activity of Bad by activating intracellular signaling pathways that result in the phosphorylation of Bad at Ser112 and Ser136.
• Phosphorylation at these sites results in the binding of Bad to 14-3-3 proteins and the inhibition of Bad binding to Bcl-2 and Bcl-xL.
• Akt has been shown to promote cell survival via its ability to phosphorylate Bad at Ser136.
• Ser112 has been shown to be the substrate in vivo and in vitro of p90RSK and mitochondria-anchored PKA.
• Phosphorylation of Ser155 by PKA plays a critical role in blocking the dimerization of Bad to Bcl-xL because of its position in the BH3 domain.
• Applications:Suitable for use in Western Blot.
• Other applications not tested.
• Recommended Dilution:Western Blot: 1:500.
• Diluted antibody in TBS, 5% BSA, 0.1% Tween-20.
• Incubate at 4°C overnight with gentle shaking
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only.
• For long-term storage, store at -20°C.
• Aliquots are stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
