Interleukin-1beta (IL-1beta), one of the major caspase-1 targets, is a multifunctional cytokine that is involved in a host of immune and proinflammatory responses.
It is produced primarily by activated monocytes and macrophages.
It signals through various adaptor proteins and kinases that lead to activation of numerous downstream targets.
Human IL-1beta is synthesized as a 31 kD precursor.
To gain activity, the precursor must be cleaved by caspase-1 between Asp116 and Ala117 to yield a 17 kD mature form.
Detection of the 17 kD mature form of IL-1beta is a good indicator of caspase-1 activity.
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:Direct ELISA: 0.15-0.30ug/ml using 100ul/well.
Allows the detection of at least 0.2ng/well
Western Blot: 0.1-0.2ug/ml.
The detection limit is 1.5-3.0ng/lane, under either reducing or non-reducing conditions.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
