Recommended Dilution:Western Blot: 1:3000 using TBA (not PBS) and blocking is best performed with highest quality IgG-free BSA.
Recognizes ~75kD band in non-acid-treated serum samples corresponding to the GDNF/GDNFR complex.
When samples are acid treated, a prominent 32kD (dimer) and weaker 16kD monomer are observed.
Acid treatment: homogenized tissue samples in PBS are acid-treated by adding 1N HCl until the pH is less than 3.0 (pH paper strips), incubated 15 minutes at room temperature and then neutralized by adding 1N NaOH to pH 7.6.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
