• This gene, CYP3A4, encodes a member of the cytochrome P450 superfamily of enzymes.
• The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism andsynthesis of cholesterol, steroids and other lipids.
• This protein localizes to the endoplasmicreticulum and its expression is induced by glucocorticoids and some pharmacological agents.
• This enzyme is involved in the metabolism of approximately half the drugs which are are used today, including acetaminophen, codeine, cyclosporin A, diazepam and erythromycin.
• The enzyme also metabolizes some steroids and carcinogens.
• This gene is part of a cluster of cytochrome P450 genes on chromosome 7q21.1.
• Previously another CYP3A gene, CYP3A3, was thought to exist; however, it is now thought that this sequence represents a transcript variant of CYP3A4.
• Applications:Suitable for use in Western Blot and Immunohistochemistry.
• Other applications have not been tested.
• Recommended Dilutions:Western Blot: 1:1000-1:2000.
• Using a microsomal membrane fractions, detects a band at ~54kD although other bands may be present as well.
• Successfully detects CYP3A4 on LICOR fluorescent westerns
• Immunohistochemistry: 1:200-1:400 on 10% formalin fixed, paraffin embedded tissues.
• Microwave-citric acid antigen recovery is suggested
• Optimal dilutions to be determined by the researcher.
• Control:Human liver microsomes
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
