特徴

• Cysteine-string proteins are membrane-bound proteins associated primarily with nerve endings and synaptic vesicles in the brain where they serve an important role in presynaptic function and may be involved in calcium-dependent neurotransmitter release at nerve endings.
• CSPs posses a strech of amino acids highly enriched in cysteine residues that are post-translationally palmitoylated.
• CSPs may function as homodimers.
• Applications:Suitable for use in Immunohistochemistry, Western Blot and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilutions:Western Blot: 1:5000 non-reducing conditionOptimal dilution determined by the researcher.
• Western Blot Technical Notes:CSPs should be resolved on a 12-15% SDS-PAGE gel under non-reducing conditions.
• They usually migrate as 33-35kD species in vertebrates (a dimer at twice this mass may also be seen).
• If one must use reducing conditions for gel samples, it is necessary to alkylate CSPs, otherwise they tend to smear.
• To ensure proper identification of CSP, the fact that CSPs show a substantial shift in electrophoretic mobility (by about 6-7kD) after deacylation (using neutral hydroxylamine (1M) or 0.1M KOH in methanol) can be used.
• Deacylation usually results in some loss of signal on immunoblot, so be sure to use enough protein.
• Alternatively, all known CSPs are quite acidic (pH about 4.6-4.8), thus 2-dimensional gels can be useful to distinguish some CSPs.
• Finally, note that some treatments cause partial deacylation of CSPs, which causes a smear of immunoreactivity to masses below that of the fatty acylated monomer.

• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
