Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases.
Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K.
Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilutions:Western Blot: 0.5-2ug/ml detects phospho-LAT (Tyr191) in RIPA lysates from Jurkat cells stimulated with C305.
Jurkat cells were incubated without or with, a monoclonal antibody that binds Jurkat T-cell receptors.
Cell lysates were prepared, resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-LAT (Tyr191) (2ug/ml), or anti- LAT (1ug/ml).
Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system
Optimal dilutions to be determined by the researcher.
