特徴

• Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes.
• Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4).
• The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures.
• This gene is intronless and encodes a member of the histone H3 family.
• Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element.
• This gene is located separately from the other H3 genes that are in the histone gene cluster on chromosome 6p22-p21.3.
• Applications:Suitable for use in Western Blot, ChIP and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilutions:Western Blot: 1:2000-1:10,000 detects methylated Histone H3 in acid extracted proteins from HeLa cells.
• 1:2000-1:5000 dilution of a previous lot detected methylated Histone H3 in acid extracted proteins from HeLa and tetrahymena cells.
• HeLa cell acid precipitate was resolved by electrophoresis, transferred to nitrocellulose and probed with a 1:5000 dilution of H5110-14J.
• Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
• Chromatin Immunoprecipitation (ChIP): 5ul immunoprecipitates methylated Histone H3 from1x10e6 3T3/A31 cells.
• Immunocytochemistry: 1:500 shows positive immunostaining for methylated Histone H3 in L6 cells fixed with 4% paraformaldehyde and permeabilized using 0.1% Triton X-100
• Optimcal dilutions should be determined by the researcher.

• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
