The action of many hormones is mediated by the generation of the intracellular second messenger cAMP.
The predominant effect of cAMP is to activate PKA (cAMP-dependent protein kinase).
Four molecules of cAMP bind each dormant PKA holoenzyme activating the kinase by releasing the catalytic (C) subunits from the regulatory (R) subunit-cAMP complex.
The cellular location of PKA is dictated by the R subunit.
Two classes of R subunit exist; RI and RII, which form the type I and type II PKA holoenzymes respectively.
The RI isoforms (RIa and RIb) are thought to be primarily cytoplasmic whereas a significant proportion of the RII isoforms (RIIa and RIIb) are particular and associate with the plasma membrane, cytoskeletal components, endoplasmic reticulum, secretory granules or nuclei.
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilutions:Western Blot: 0.5-2ug/ml detected PKA, NT (42kD) in both rat brain microsomal preparation and PC-12 RIPA cell lysates.
Rat brain preparation was resolved by electrophoresis, transferred to nitrocellulose and probed with P9102-91A (0.5ug/ml).
Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system

Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
