Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases.
Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K.
Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells
Applications:Suitable for use in Western Blot and Immunoprecipitation.
Other applications have not been tested.
Recommended Dilutions:Western Blot: 2-4ug/ml detects LAT in RIPA lysates from Jurkat cells.
Jurkat cell lysate was resolved by electrophoresis, transfered to PVDF and probed with L1390 (2ug/ml).
Proteins were visualized using IgG (HRP) goat anti-rabbit secondary antibody and a chemiluminescence detection system
Immunoprecipitation: 8-10ug immunoprecipitated LAT from 500ug of Jurkat RIPA lysateOptimal dilutions to be determined by the researcher.
Included Positive Antigen Control:J9000: Jurkat lysate.
Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation.
Load 20ug of reduced lysate per lane for minigels.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
