The tumor suppressor p53 is a critical mediator of cellular responses to DNA damage.
p53 stability and transcriptional activity are modulated by post-translational modifications such as phosphorylation and acetylation.
In response to UV or gamma irradiation, the p53 tetramerization domain becomes a target for acetylation by the transcription factors CREB binding protein (CBP)/p300 and PCAF, which also function as co-activators for p53-mediated transcriptional activation.
p300, which possesses intrinsic acetyl transferase activity, specifically acetylates p53 on lysine residue 373 and possibly lysine 382.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:250-1:500; Detects p53-GST acetylated by recombinant PCAF, Active (Cat# P3114) in an in vitro acetyl transferase assay.
In vitro acetyl transferase reaction products were resolved by electrophoresis, transferred to nitrocellulose and probed with P1001-30C.
Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP* and a chemiluminescence detection system.
Note: The molecular weight of GST is 25.5kD.
Consequently, the p53-GST fusion protein is detected at ~80kD
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
