The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle.
Cyclins function as regulators of CDK kinases.
Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event.
In contrast to cyclin A1, which is present only in germ cells, this cyclin is expressed in all tissues tested.
This cyclin binds and activates CDC2 or CDK2 kinases, and thus promotes both cell cycle G1/S and G2/M transitions.
Applications:Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 0.5-2ug/ml detects p58 Cyclin A in HeLa nuclear extract.
HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with 2ug/ml C8452-08.
Proteins were visualized using an IgG (HRP) goat anti-rabbit secondary antibody and a chemiluminescence detection system
Immunoprecipitation: 1-5ug/ml.
The precipitate may be used to perform a histone H1 kinase assay
Immunocytochemistry: 10ug/mlOptimal dilution to be determined by the researcher.
Positive Control Included:H1838: HeLa nuclear extract.
Add an equal volume of Laemmli reducing sample buffer to 5-10ul of extract and boil for 5 minutes to reduce the preparation.
Load 10-20ug of reduced lysate per lane for minigels.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile dH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
