• The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle.
• Cyclins function as regulators of CDK kinases.
• Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event.
• In contrast to cyclin A1, which is present only in germ cells, this cyclin is expressed in all tissues tested.
• This cyclin binds and activates CDC2 or CDK2 kinases, and thus promotes both cell cycle G1/S and G2/M transitions.
• Applications:Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilutions:Western Blot: 0.5-2ug/ml detects p58 Cyclin A in HeLa nuclear extract.
• HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with 2ug/ml C8452-08.
• Proteins were visualized using an IgG (HRP) goat anti-rabbit secondary antibody and a chemiluminescence detection system
• Immunoprecipitation: 1-5ug/ml.
• The precipitate may be used to perform a histone H1 kinase assay
• Immunocytochemistry: 10ug/mlOptimal dilution to be determined by the researcher.
• Positive Control Included:H1838: HeLa nuclear extract.
• Add an equal volume of Laemmli reducing sample buffer to 5-10ul of extract and boil for 5 minutes to reduce the preparation.
• Load 10-20ug of reduced lysate per lane for minigels.
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
• Reconstitute with sterile dH2O.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
