Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts.
TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions.
Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis.
Applications:Suitable for use in Neutralization, ELISA and Western Blot.
Other applications have not been tested.
Recommended Dilution:Neutralization: 0.07-0.1ug/ml will yield one-half maximal inhibition [ND50] of the biological activity (2.0ng/ml)
ELISA: 0.5-2ug/ml is detection by direct ELISA (using 100ul/well antibody solution).
In conjunction with compatible secondary reagents this antibody allows detection of 0.2-0.4ng/well of recombinant rat TNF-a
Western Blot: 0.1-0.2ug/ml.
Used in conjunction with compatible secondary reagents the detection limit is 1.5-3.0ng/lane, under either reducing or non-reducing conditions.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
