NFkB exists in the cytoplasm as a complex with a 38kD inhibitor protein, IkB (MAD-3).
Activation of NFkB by different stimuli, including TNFa and PMA, requires the dissociation of the inhibitor protein.
Dissociation is regulated by the phosphorylation of IkB on serine residues 32 and 36 which targets IkB for ubiquinitation and proteasome mediated degradation.
The NFkB complex then translocates to the nucleus and activates transcription of responsive genes.
Applications:Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:ELISA: 1:1000-5000Western Blot: 1:500-1000 Optimal dilutions to be determine by researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C or colder.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
