The interaction between CD44 and hyaluronan may mediate binding of cells to basement membranes.
Several CD44 isoforms or variants arise from alternative splicing of mRNA transcribed from a single gene located on chromosome 11.
Most cell types express the st andard CD44, while the expression of the isoforms resulting from the alternative splicing is of limited distribution.
The CD44 st andard form is observed to associate with Src and the Src family members Fyn and Yes.
Stimulation of CD44 st andard expressed by SK-OV-3.ipl ovarian cancer cells by the addition of hyaluronan results in recruitment of Src to a CD44-containing complex and increases phosphorylation of Src at tyrosine 418.
Phosphorylation at this residue increases Src kinase activity.
Cortactin, a cytoskeleton-associated protein and a known substrate for Src, is also phosphorylated in response to CD44 stimulation with hyaluronan and this phosphorylation reduces cortactin's ability to cross-link filamentous actin (Bourguignon et al.
2001).
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
