• Microtubule-associated protein tau (UniProt: P10636; also known as Neurofibrillary tangle protein Paired helical filament-tau (PHF-tau)) is encoded by the MAPT (also known as MAPTL, MTBT1, TAU) gene (Gene ID: 4137) in human.
• Tau is a highly abundant protein found predominantly in the nervous system.
• Its primary role is to stabilize and support microtubule assembly, which are essential for maintaining the structure and function of nerve cells.
• It is present throughout the brain, particularly in regions involved in memory and cognition.
• MAPT gene consists of multiple exons that undergo alternative splicing, resulting in the generation of different isoforms that differ in the number and arrangement of repeats in the microtubule-binding domain.
• The expression of the MAPT transcripts varies and depends on the stage of neuronal maturation and neuron type.
• Aberrant aggregation and accumulation of tau protein have been linked to several neurodegenerative diseases collectively known as tauopathies.
• The most common tauopathy is Alzheimer′s disease, where hyperphosphorylated tau protein forms neurofibrillary tangles inside neurons.
• Other tauopathies include frontotemporal dementia, progressive supranuclear palsy, and cortico-basal degeneration.
• These diseases are characterized by cognitive decline, motor disturbances, and memory loss.
