The tumor suppressor protein p53 is a key element of intracellular anticancer protection.
It mediates cell cycle arrest or apoptosis in response to DNA damage or to starvation for pyrimidine nukleotides.
It is up-regulated in response to these stress signals and stimulated to activate transcription of specific genes, resulting in expression of p21waf1 and other proteins involved in G1 or G2/M arrest, or proteins that trigger apoptosis, such as Bcl-2.
The structure of p53 comprises N-terminal transactivation domain, central DNA-binding domain, oligomerization domain, and C-terminal regulatory domain.
There are various phosphorylation sites on p53, of which the phosphorylation at Ser15 is important for p53 activation and stabilization.
Applications:Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry, Flow Cytometry and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:Flow Cytometry: 1-5ug/ml.
Intracellular staining
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4℃ if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20℃.
Aliquots are stable at -20℃ for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
