• Lethal(2) giant larvae protein homolog 1 (UniProt: Q80Y17; also known as LLGL, Mgl-1, Mlgl) is encoded by the Llgl1 (also known as Llglh) gene (Gene ID: 16897) in murine species.
• LLGL belongs to the WD repeat L(2)GL family.
• LLGL is a cortical cytoskeleton protein that is found in a complex involved in maintenance of cell polarity and epithelial integrity.
• It is involved in the regulation of mitotic spindle orientation, proliferation, differentiation, and tissue organization of neuroepithelial cells.
• It is generally enriched in developing axons.
• LLGL stability and function is achieve by its interaction with RanBPM protein, which primarily acts as a scaffolding protein.
• Its N-terminal region containing SPRY domain is shown to be responsible for binding to RanBPM and this binding functionally extends its half-life by preventing its turnover through ubiquitin-proteasomal pathway.
• RanBPM is shown enhance tumor suppressor activity of LLGL by inhibiting LLGL-mediated cell proliferation and cell migration.
