Progranulin is a ubiquitously expressed, 88kD, secreted glycoprotein.
Structurally, it does not belong to any of the well established growth factor families.
Human Progranulin is 593 amino acids (aa) in length and contains a 17aa signal sequence and 5 potential sites for N-linked glycosylation.
Progranulin is involved in the regulation of cellular proliferation, as well as differentiation, development and pathological processes.
It has been isolated as a differentially expressed gene during mesothelial differentiation, macrophage development, the development of rheumatoid arthritis and osteoarthritis, sexual differentiation of the brain and has also been shown to be a mediator of cartilage proliferation and of wound response and tissue repair.
High levels of Progranulin expression have been found to be associated with several human cancers and are believed to contribute to tumorigenesis in breast cancer, clear cell renal carcinoma, invasive ovarian carcinoma, glioblastoma, adipocyte teratoma and multiple myeloma.
In addition, mutations in the Progranulin gene are a cause of frontotemporal dementia, and increased expression of Progranulin is seen in activated microglia in many neurodegenerative diseases including Creutzfeldt-Jakob disease, motor neuron disease and Alzheimer's disease.
Mutations in Progranulin causing neuro- degenerative disease indicate that Progranulin is important for neuronal survival.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 2ug/ml detects HepG2 human hepatocellular carcinoma cell line and mouse liver tissue under reducing conditionsImmunoprecipitation: 5-25ug/ml in in immersion fixed paraffin-embedded sections of human breast cancer tissueOptimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20℃.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20℃.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
