• Laminins, a family of extracellular matrix glycoproteins, are the major noncollagenous constituent of basement membranes.
• They have been implicated in a wide variety of biological processes including cell adhesion, differentiation, migration, signaling, neurite outgrowth and metastasis.
• Variations in the expression of laminins have been observed in embryogenesis, organogenesis, post traumatic healing and cancer.
• Laminins have been provoked by the discovery of their ability to promote neurite regeneration.
• Laminins are composed of 3 non identical chains: laminin alpha, beta and gamma (formerly A, B1, and B2, respectively).
• The gamma 1 chain, formerly thought to be a beta chain, contains structural domains similar to beta chains, however, lacks the short alpha region separating domains I and II.
• Embryos of transgenic mice in which both alleles of the gamma 1 chain gene were inactivated by homologous recombination, lacked basement membranes, indicating that laminin, gamma 1 chain is necessary for laminin heterotrimer assembly.
• Applications:Suitable for use in Immunohistochemistry.
• Other applications have not been tested.
• Recommended Dilutions:Immunohistochemistry (formalin-fixed, paraffin-embedded): 1:50-1:500 detected strong membrane staining of basement membrane of formalin-fixed, paraffin-embedded human tumor cells.
• Pretreat tissues using a citrate buffer, pH 6.0 and high heat epitope retrieval techniques
• Optimal dilutions to be determined by the researcher.
• Positive Control:Tubular carcinoma of the colon and fibroneuroma
• Storage and Stability:May be stored at 4℃ for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20℃.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
