特徴

• Recombinant monoclonal antibody to TIGIT with variable regions (i.e., specificity) from the hybridoma 1B4.
• This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
• The T cell immunoreceptor with Ig and ITIM domains (TIGIT) is a member of the PVR (poliovirus receptor) family of immunoglobin proteins.
• It is expressed on several classes of T cells including follicular B helper T cells (TFH).
• TIGIT has been shown to bind PVR with high affinity; this binding is thought to assist interactions between TFH and dendritic cells to regulate T cell dependent B cell responses.
• TIGIT is upregulated on exhausted T cells in chronic viral infections and cancer.
• Blockade of both TIGIT and PD-1 pathways leads to tumor rejection in mice suggesting that it may be of therapeutic use against cancer.
• Applications:Suitable for use in ELISA, Blocking and Flow Cytometry.
• Other applications not tested.
• Application Notes:The binding specificity of this antibody for TIGIT has been confirmed in ELISA analysis and by flow cytometry using P815 cells transfected with mouse TIGIT and activated mouse splenocytes (Dixon et al, 2018).
• This antibody also specifically stains activated primary wild-type T cells (Dixon et al, 2018).
• This antibody fully blocks CD155 binding to TIGIT, and acts as a TIGIT blocking antibody in vivo.
• Administration of this antibody increases experimental autoimmune encephalomyelitis (EAE) severity, and retards the growth of established MC38 colon carcinoma and GL261 glioblastoma (Dixon et al, 2018).
• This antibody synergizes with anti-PD-1 blocking antibodies to achieve complete tumor regression in colon carcinoma models (Dixon et al, 2018).
• The original mouse IgG1 antibody does not deplete TIGIT-expressing CD4+ or CD8+ T cells in vivo, nor affect T cell proliferation in vitro (Dixon et al, 2018).
• Recommended Dilution:Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4℃ for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20℃.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
