PARP [Poly(ADP-ribose) Polymerase], also known as ADPRT and PPOL, is a 118kD enzyme that uses NAD as a substrate to catalyze the covalent transfer of ADP-ribose to a variety of nuclear protein acceptors.
ADP ribosyltransferase is required for cellular repair, and PARP expression is induced by single-strand breaks in DNA.
PARP is proteolytically cleaved by Caspase-3 into two fragments of 89- and 24-kD in one of the hallmark events of apoptosis.
Applications:Suitable for use in ELISA, Western Blot Immunoprecipitation, Flow Cytometry and immunocytochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000Immunohistochemistry: 1-15ug/ml using immersion fixed paraffin-embedded sections of human kidneyImmunocytochemistry: 3-25ug/ml using immersion fixed 786-O human renal cell adenocarcinoma cell lineOptimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4℃.
For long-term storage, aliquot and store at 4℃.
Do not freeze.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Further dilutions can be made in assay buffer.
