• The protein is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes.
• This cytokine has pleiotropic effects in immunoregulation and inflammation.
• It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages.
• It also enhances B cell survival, proliferation, and antibody production.
• This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway.
• Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract.
• Applications:Suitable for use in ELISA, Western Blot and Neutralization.
• Other applications not tested.
• Recommended Dilution:ELISA: In a sandwich ELISA (assuming 100ul/well), a concentration of 4-8ug/ml of this antibody will detect at least 1600pg/ml of recombinant murine IL-10 when used with biotinylated antigen affinity purified anti-murine IL-10 as the detection antibody at a concentration of approximately 0.5-1ug/ml
• Western Blot: To detect mIL-10 by Western Blot analysis this antibody can be used at a concentration of 1-2ug/ml.
• Used in conjunction with compatible secondary reagents the detection limit for recombinant mIL-10 is 2-4ng/lane, under reducing or non-reducing conditions
• Neutralization: To yield one-half maximal inhibition of the biological activity of Murine IL-10 (0.5ng/ml), a concentration of 3-5ug/ml of this antibody is required
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20℃.
• Reconstitute with sterile ddH2O.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20℃.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
