The protein is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes.
This cytokine has pleiotropic effects in immunoregulation and inflammation.
It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages.
It also enhances B cell survival, proliferation, and antibody production.
This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway.
Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract.
Applications:Suitable for use in ELISA, Western Blot and Neutralization.
Other applications not tested.
Recommended Dilution:ELISA: In a sandwich ELISA (assuming 100ul/well), a concentration of 4-8ug/ml of this antibody will detect at least 1600pg/ml of recombinant murine IL-10 when used with biotinylated antigen affinity purified anti-murine IL-10 as the detection antibody at a concentration of approximately 0.5-1ug/ml
Western Blot: To detect mIL-10 by Western Blot analysis this antibody can be used at a concentration of 1-2ug/ml.
Used in conjunction with compatible secondary reagents the detection limit for recombinant mIL-10 is 2-4ng/lane, under reducing or non-reducing conditions
Neutralization: To yield one-half maximal inhibition of the biological activity of Murine IL-10 (0.5ng/ml), a concentration of 3-5ug/ml of this antibody is required
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20℃.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20℃.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
