概要:3T3-L1 is a continuous subclone of the 3T3-Swiss Albino cell line developed through clonal isolation.
These cells have an unstable karyotype with 2n = 40 chromosomes and are susceptible to murine leukemia virus, murine sarcoma virus, vesicular stomatitis, vaccinia, herpes simplex, and N-tropic oncornaviruses C.
These cells undergo adipocyte differentiation when the culture reaches confluence, and the cells enter a resting state.
During this process, a high serum content in the medium enhances fat accumulation.
The appearance of mature adipocytes can take weeks to achieve.
These cells accumulate large amounts of triglyceride fat when in a resting state, and lipolytic agents can reduce the accumulation.
Continued proliferation can revert lipid accumulation, eliminating most of the lipids.
3T3-L1 cells are widely used in applications such as adipogenesis and obesity research since the 3T3-L1 preadipocyte is a good model for studying the molecular events that differentiate fibroblasts into mature adipocytes, including transcriptional regulation of adipocyte-specific genes, intracellular signalling pathways, and growth factors and hormones.
These cells are a suitable transfection host and can be used to screen drugs that determine their effect on cell fat accumulation.
Insulin is expressed, and triglycerides are among the genes expressed in these cells.
3T3-L1 is also specialized for collagen synthesis.
Before its adipose conversion, it makes about as much collagen as other 3T3 cells.
