• 背景：Vinculin is a cytoskeletal protein that is a member of the vinculin/α-Catenin family. Vinculin is allocated mainly at cell-cell junctions and cell-extracellular matrix （ECM） adhesion. Vinculin is an actin-binding protein that detects the mechanical properties of the extracellular environment. It plays an important role in coupling, transmiting, transduceing, and regulating mechanical force between the cytoskeleton and adhesion receptors. Vinculin can form two structural states, an open （active） and closed （inactive） state, decided by conformational interaction between its head and tail domains. Vinculin also takes part in the mechano-chemical signal transmission of cells by binding to a variety of focal adhesion or cytoskeletal proteins, and is essential in cell's adhesion, extension, motion, proliferation and survival.
